چکیده
Cremastra appendiculata var. variabilis (Blume) I.D. Lund), also known as single-leaf cremastra (SLC), is a rare and threatened species native to Korea, it has the potential be grown beautiful flowering pot or garden plant. There still no reliable strategy multiply SLC. Thus, an effective method for propagating was needed its conservation mass production. In present study, we examined effects of culture media, seed age, activated charcoal, plant growth regulators on in vitro asymbiotic germination, secondary protocorm induction, seedling formation. Asymbiotic germination SLC influenced by their interaction. The addition charcoal (500 mg/L) Murashige Skoog (MS) medium increased rate germination. seeds were best germinated (91.9%) culturing MS supplemented with mg/L), α-naphthaleneacetic acid (3 µM), kinetin (1 µM). highest number (28.9) protocorms produced when cultured containing 6-benzyladenine (4 µM) (2 When cultivated gibberellic they able transform into success (78.7%). propagation protocol described here may helpful restoration programs large-scale

In vitro asymbiotic germination of immature seed and formation of protocorm by Cephalanthera falcata (Orchidaceae).
BACKGROUND AND AIMS Many Orchidaceous species are threatened globally by development and over-collection from their natural habitats for horticultural purposes. Artificial propagation from seeds is difficult in most terrestrial orchids native to temperate regions. Seed production is another limiting factor in the artificial propagation for these species because of the lessened probability of po...
متن کاملFive New Biphenanthrenes from Cremastra appendiculata.
Five new biphenanthrenes, cremaphenanthrenes A-E (1-5), along with six known ones, were isolated from the ethanolic extract of the tubers of Cremastra appendiculata (D. Don) Makino (Orchidaceae). Their structures were elucidated on the basis of extensive spectroscopic analyses. All the compounds obtained were tested in vitro for cytotoxic activities against colon (HCT-116), cervix (Hela), and b...
متن کاملAn Effective Nutrient Media for Asymbiotic Seed Germination and In Vitro Seedling Development of Phalaenopsis ‘Bahia Blanca’
Our study was to develop a rapid and successful method for promoting asymbiotic seed germination and growth and development of protocorm like bodies (PLBs) and subsequent plantlets of Phalaenopsis orchid. Four basal media i.e. Murashige and Skoog (MS), half strength MS (½MS), Knudson C (KC) and, Vacin and Went (VW) supplemented with coconut water and peptone were examined for their effectivenes...
متن کاملEx situ conservation of Cymbidium eburneum Lindl.: a threatened and vulnerable orchid, by asymbiotic seed germination
The population of many splendid orchids is reducing from their natural habitats at an alarming rate and their conservation is becoming a matter of global concern. Asymbiotic seed germination has been applied for ex situ conservation of rare, endangered and threatened orchid taxa and could provide rapid means their multiplication. In the present study reported here, seeds of an epiphytic and rar...
متن کاملEmbryo development in association with asymbiotic seed germination in vitro of Paphiopedilum armeniacum S. C. Chen et F. Y. Liu
This paper documents the key anatomical features during the development of P. armeniacum zygotic embryos and their ability to germinate asymbiotically in vitro. This study also examines the effect of media and seed pretreatments on seed germination and subsequent seedling growth. Seeds collected from pods 45 days after pollination (DAP) did not germinate while 95 DAP seeds displayed the highest...
